Life Science

The automation of applications in life sciences such as proteomics studies, cancer and biomarker studies is very valuable. Typical are the large sample series from cohorts of patients requiring highly reproducible processing, a particular strength of the PAL System.

Temperature controlled sample storage, overlapping and prep-ahead sample preparation, assure that each sample is processed exactly the same way to achieve good reproducibility and comparability of the data. Unattended, fully automated operation of the PAL System is integrated into the chromatographic data systems of all leading manufacturers.

PAL Systems automate a wide range of sample preparation applications in clinical, biotechnological, and biomedical research using dedicated tools and modules with efficient workflows for LC/MS and GC/MS analysis.

Applications include the Bligh & Dyer method, liquid/liquid extraction, protein precipitation and centrifugation steps. For metabolomic profiling the derivatization of analytes is required for high sensitivity analysis.

For therapeutic drug monitoring a dedicated PAL LC/MS Tool offers the direct sampling from primary tubes. An optical sensor is positioned on the sample loop of the tool which monitors the aspiration of blood or serum sample, air gap or bubbles.

Breath gas analysis for volatiles in clinical applications for diagnosis and cancer biomarker research is another fast growing area of Solid Phase Micro Extraction (SPME) using the rugged SPME Arrows.

The PAL System supports research for the biological activity of proteins by elucidation of their 3-dimensional structure in its active or inactive states. Hydrogen Deuterium Exchange (HDX) is the labeling technique which allows the study of how protein chains are folded during the interaction with their binding partners under physiological conditions.

Automated Parallel Derivatization of Metabolites

For metabolite profiling chemical derivatization has been used to improve MS sensitivity and LC retention. However, for multi-analytes quantification, the number of available isotopically labelled internal standards is limited. And, there is no single workflow which can provide large-scale metabolomics coverage in particular for polar metabolites.

A single-sample workflow for joint metabolomic and proteomic analysis ofclinical specimens

Integrating proteomic and metabolomic data enhances ourunderstanding of biological processes and diseasemechanisms. Traditional multi-omics workflows often requireseparate sample processing, leading to increased variability. To address this, we developed MTBE-SP3, an optimizedsingle-sample workflow that combines a 75% EtOH/MTBEmetabolite extraction with automated single-pot solid-phase-enhanced sample preparation for proteins.
